Human Cell Line Identity Verification

Sample Preparation

DNA is isolated by the investigator, who submits the appropriate quantity and concentration of DNA to the core. 

DNA isolation - The MDL has assessed the Promega STR profiling systems with genomic DNA isolated by several Qiagen  DNA isolation kits. Assure that your method of DNA isolation and solubilization produces samples that are compatible with PCR. The core laboratory will not purify submitted DNA that contains inhibitors of PCR.

DNA quantity and concentration - A minimum of 500 ng of DNA at a concentration of 50 ng/ul
​(10 ul total sample volume ) is required for the genotyping assay and for spectrophotometric quality control determinations.

Sample Submission

Consultation - If it is helpful, you may arrange for a consultation with the lab director by email (sigitas_verselis@dfci.harvard.edu) to discuss any questions that you may have.

Sample Submission - Samples should be submitted as soon as they are prepared to get in queue for the next batch of analyses. Generally, samples are analyzed twice each month, usually the middle and end of each month.

Application for Services - An "Application for services" form is available for downloading and must be completed. Please ensure that all fields on the application are filled in. The MDL will not process samples if information is missing. Comment fields and the embedded "project description" document are optional.

Please forward an electronic copy of the completed form by email to the lab director, Sig Verselis Ph.D. 
Include a printed copy of the completed form with your samples when dropping them off.

Sample drop-off - The Local Sample Drop-off room is located on the main campus of the Dana-Farber in the "Fish Room" near the 3rd floor bridge of the Smith Building leading to the Jimmy Fund Building. Place DNA samples in 1.5mL microcentrifuge tubes (with the sample name written on the top, and your initials written on the side) along with your printed application form, into a zip lock plastic bag (available in the Fish Room) and store on the top shelf in the +4°C refrigerator with glass doors.

You will need a Dana-Farber ID card to enter the room. If you do not have one, please go to the Security desk in the lobby of the Smith or Dana buildings and request to be escorted to the "fish room". The phone number for security is 617-632-3131 (extension 2-3131 within DFCI).

If the local drop-off location is not accessible, please contact the lab director by email 
(sigitas_verselis@dfci.harvard.edu) to make alternate arrangements for sample delivery.
