miRNA Expression Profiling - Step 2: cDNA synthesis

Sixty nanograms of submitted RNA is used in the reverse transcription reaction. A stem-loop primer for each target miRNA is present in the reaction. The 3' region of each primer is complementary to the 3' region of its target miRNA. The 5' region of the primer is a non-miRNA "tail" and contains a sequence that will serve as the reverse primer in the pre-amplification and TaqMan® reactions.
